Once-daily sertraline, with a half-life of 25 to 26 hours, 10 is an example of a medication with a substantial pharmacokinetic buffer against inaccurate pill splitting. A detailed cost analysis of the 48 remaining medications using data from the available pharmacy claims records allowed us to determine actual cost, current rates of pill splitting among MGH physicians, and potential savings from extended use of this strategy. Eliminating those medications with minimal usage in the MGH population, we identified 11 recommended medications for which pill splitting is clinically appropriate and cost saving. To calculate current rates of pill splitting for these medications, we used the following methods: for each daily dose of each medication, we calculated the proportion of prescriptions for which 2-to-1 splitting was implied by the number of pills provided and the days of therapy supplied by the prescription. For example, for all patients prescribed lisinopril 10 mg per day, we compared the number achieving this dose via 10-mg tablets 30 tablets provided for 30 days ; with the number achieving this dose via 20-mg tablets split 2-to-1 15 tablets provided for 30 days ; . For each medication, we reported the aggregate rate of pill splitting across all possible 2-to-1 splitting possibilities. During our investigation, no organizational efforts were in place to promote pill splitting. Olanzapine concentrations as mu ch 58% greater than those who do, whereas females who do not smoke may have concentrations as mu ch 93% higher than those who do. Risperidone No studies have been able to document a correlation between risperidone and its active 9-hydroxy metabolite and therapeutic response. A weak relationship between risperidone and the daily risperidone dose milligrams day ; has been reported, although a more robust relationship between total risperidone plasma concentration risperidone + 9hydroxy risperidone ; and dose r2 0.50, N 280 ; was observed.36 This finding suggested that studies pursuing SCTER for risperidone should correlate the total active drug concentration with the rating scale change scores. One study using this strategy was unable to discern a significant correlation between the total risperidone concentration or daily risperidone dose and an adverse effect scale.37 One additional point of clinical interest in this study was that of the 22 patients administered the clinically optimum dose of 6 mg day, 90% had total risperidone concentrations between 50150 nmol L 2162 ng mL and ondansetron.

Among these, estrogens have a significant impact on bone metabolism. Estrogens receptors ER et ER ; are expressed by osteoblasts from normal and pathological osteoporosis ; cells, but their expression in scoliotic cells and tissues was not demonstrated yet. The objective of this project is to ascertain the expression of the estrogen receptors ER and ER and to analyse polymorphisms of the ER gene in adolescent idiopathic scoliosis patients AIS ; . This research aims to clarify the role of estrogens and estrogen receptors in the pathogenesis of AIS. Methodology : Using RT-PCR, we compared ER and ER mRNA expression in osteoblasts from control and AIS patients n 40 ; . Through use of Western blot, we identified the two known E2 receptors ER and ER ; in osteosblastic cell lines and osteoblasts from scoliotic patients. Polymorphisms of the first intron of the ER gene, which contains the XbaI and PvuII polymorphisms, were investigated by PCR following by digestion with restriction enzyme and using the genomic DNA from lymphocytes isolated from scoliotic patients n 33 ; . Results: We observed a decrease of ER expression in some osteoblasts from scoliotic patients. The XbaI and PvuII polymorphisms were found in 70% 23 33 ; and 80% 26 33 ; of the cases respectively. Of the 33 cases, 21 presented both digestion sites. Western blot analysis allowed us to detect ER and observe the induction of ER by osteoblasts from scoliotic patients. Conclusion : The expression of ER appears altered in a number of AIS patients. The XbaI and PvuII polymorphisms are represented in the AIS population. The functional impact of these polymorphisms remains to be determined. This research could help in the identification of prognostic tools for the prediction of the development of AIS and assist in the development of new molecular strategies to prevent or reduce spine deformity. The importance of this proposal is in its explicit goal of clearly establishing the specific role of estrogens and their receptors in pathogenic mechanisms involved in AIS. New York investor Wilbur Ross may have hit some snags on the way to rolling up the auto interiors sector. Collins & Aikman Corp., a linchpin of Ross's acquisition strategy, is looking increasingly like a company that plans to emerge from Chapter 11 bankruptcy protection as a stand-alone entity rather than being broken up and sold. CEO Frank Macher is trying to convince creditors that there's value in having the company exit bankruptcy intact. If he can, Ross will have missed a key quarry in Macher his effort to build an interiors empire in North America. Lear Corp. also would lose under that scenario. The seat and interiors supplier in October told of plans to put its money-losing interior-trim business in a joint venture with Ross. But if Ross cannot land Collins & Aikman, that deal may fall apart. Lear CEO Robert Rossiter told auto analysts last month that he believed a possible Lear deal with Ross depended on Ross completing the acquisition of Collins & Aikman. "The way we have it structured right now, the deal probably wouldn't happen without the acquisition of Collins & Aikman, " Rossiter said at a meeting of the Auto Analysts of New York in January in Dearborn. Without Ross and the joint venture, the future of Lear's interiortrim business is uncertain. That business helped drag down the company's earnings last year. On Jan. 25, rating service Standard & Poor's put the company's debt ratings on CreditWatch with negative implications because of concerns over earnings and cash. Ross creates healthy companies by buying assets after bankruptcy court has eliminated debt and legacy costs. This bargain-basement strategy helped him consolidate an insolvent steel industry. Last November, the Ross-Lear joint venture, along with financier Franklin Mutual Advisers L.L.C., said it would acquire Collins & Aikman's European operations for $600 million. But buying Collins & Aikman's North American operations may prove more difficult. Lear spokesman Mel Stephens declined comment. Lear canceled an interview scheduled Feb. 2 between Automotive News and Lear CFO Dave Wajsgras. Ross says he plans to stay the course. The first national Medicinal Chemistry Symposium I sponsored by the Division ; was held in 1948 at the University of Michigan, with F.P. Blicke as general chair and Glenn E. Ullyot as program chair. Symposia have subsequently been held regularly in alternate even-numbered years, always on a university campus. In 1968 the symposium assumed an international flavor, meeting at Laval University in Quebec under joint sponsorship with the newly formed Medicinal Chemistry Division of the Chemical Institute of Canada.
